Introduction: Age-related cataract is the leading cause of blindness in world. (AP)-endonuclease APE1 and X-ray cross-complementing group 1 (XRCC1) are key enzymes taking part repair DNA damage. The association AP endonuclease-1 (APE1) plays a critical role as polymorphisms genes decreasing ability to damage, leaving human body greatly increased susceptibility cancer or age-related diseases.
 Aim objective: To study Molecular characterization its Association (APE) (XRCC1), Repair protein from patients with risk at Tertiary Care Centre, Uttar Pradesh.
 Material methods: This was case control carried out Department Anatomy & Ophthalmology RMCH&RC for period 2 year i.e., June 2020 2022. A total 500 clinical suspected cases were included our which 250 confirmed positive whereas Controls. Demographic profiles recorded after duly obtaining consent. 5ml venous blood collected Ethylene diamine tetraacetic acid tubes.

 Results: In present maximum number reported Females compared Males age 51-60 years being affected most. Expression (AP) –endonuclease more 68.4 % 62.8 %. increase expression indicates presence these damage.
 Conclusion: 68.4% 62.8%. So, there should be early screening molecular detection gene, will help clinician diagnosis well treatment cataract.
